چکیده
background: leishmania major is an intracellular parasite transmitted through the bite of the female phlebotomine sand flies. leishmania major is able to escape the host immune defense and survive within macrophages. modulation of the nf-κb (nuclear factor-kappa b) activation and suppression of the pro-inflammatory cytokines by l. major are the main evasion mechanisms that remain to be explored. this study aims to examine the expression level of the monarch-1 in l. major- infected macrophages, as a negative regulator of the nf-κb activation. methods: murine macrophage cell line (j774 a.1) was infected by metacyclic form of leishmania promastigotes at macrophage /parasite ratio of 1:10. after harvesting infected cells at different times, total rna was extracted and converted to cdna. semi-quantitative rt-pcr was performed for monarch-1 by specific primers. hypoxanthine phospho-ribosyl transferase (hprt) was used as an internal control to adjust the amount of mrna in each sample. results: semiquantitive analysis of monarch-1 mrna expression level showed a significant expres-sion increase within 6 to 30 hours after l. major infection of macrophages when compared to the control macrophages. conclusion: monarch-1 expression level reveals a significant increase in the early phase of macro-phage infection with l. major, which in turn may suppress il-12 production in leishmania infected macrophages and deeply influence the relationship between host and parasite.

Background Cancer origin is closely linked with mutations and changes in the chromosome structure, making important such cytogenetics studies [1]. Macrophage murine cell line J774-1 is derived from a reticulum cell sarcoma and presents various characteristics of primary macrophages as synthesis and lysozyme secretion, phagocytosis, Fc receptor expression [2,3]. This work aimed to analyze the fu...
متن کاملStreptococcus suis interactions with the murine macrophage cell line J774: adhesion and cytotoxicity.
Streptococcus suis capsular type 2 is an important etiological agent of swine meningitis, and it is also a zoonotic agent. Since one hypothesis of the pathogenesis of S. suis infection is that bacteria enter the bloodstream and invade the meninges and other tissues in close association with mononuclear phagocytes, the objective of the present study was to evaluate the capacity of S. suis type 2...

The Mono Mac 6 (MM6) human monocytic cell line was evaluated with the established J774 murine macrophage cell line to ascertain its effectiveness in determining the intracellular activities of antimycobacterial drugs. Cells were infected with Mycobacterium tuberculosis H37Ra and treated with drug concentrations corresponding to the MICs, as well as to threefold higher than and threefold less th...

OBJECTIVES The objective of this study was to isolate a Brucella lon mutant and to analyze the cytokine response of B. lon mutant during macrophage infection. METHODS A wild-type Brucella abortus strain was mutagenized by Tn5 transposition. From the mouse macrophage J774.A1 cells, total RNA was isolated at 0 hours, 6 hours, 12 hours, and 24 hours after infection with Brucella. Using mouse cyt...
متن کاملMechanism of lipopolysaccharide-triggered junB activation in a mouse macrophage-like cell line (J774).
The effect of lipopolysaccharide (LPS) on the activation of junB in a mouse macrophage cell line (J774) was investigated. J774 cells responded to either phorbol 12-myristate 13-acetate (PMA) or LPS by the transient increase in the expression levels of c-jun and junB mRNA, but not of junD mRNA. The prior depletion of protein kinase C from J774 cells blocked the action of PMA, but not of LPS, to ...
